Cause of Avandia Side Effects Identified; Therapy in Development

Researchers are starting to pinpoint the cause of adverse reactions seen in some diabetes patients treated with Avandia (rosiglitazone) and drugs of its class (thiazolidinediones). 
 
While they are very effective at lowering blood glucose, they can lead to fluid retention, swelling, heart complications, and even visual problems if the retina is affected by excess fluid.  
 
These effects were found to mirror the effect that diabetes itself has on the body. The exact protein (enzyme) causing this problem has been pinpointed, and an inhibitor drug is in development that will allow diabetics who also have heart disease or other risk factors to benefit from Avandia-type drugs.

Used by several million people worldwide, rosiglitazone (RSG) is an oral agent that helps patients with type 2 diabetes maintain good blood glucose levels by improving how their bodies use insulin.
 
But RSG, like all the other thiazolidinedione (TZD) drugs that can lower blood glucose levels, can cause fluid retention (edema), a condition that puts patients at greater risk for weight gain, vascular complications and heart failure. Recently, the U.S. Food and Drug Administration and GlaxoSmithKline, which manufactures the drugs, reported cases of new onset or worsening macular edema (an eye disorder that leads to blurred or distorted vision) among patients who took RSG. While reports of these complications remain rare, GlaxoSmithKline has added a warning about the risks to the drugs' labels.

Tomorrow will be one year to the day that I lost my hysband. I have done much research and feel avandia killed my husband. This has been a very difficult time for me and I would more than anything have at least tried to avenge my husband's death. Before he was put on avandia he was hale and hearty. After i i/2 years on this medicine he blew up with fluid. 27 liters of fluid tapped from his stomach. Fluid in his lungs and the kidneys, liver and heart were afected. $92.000 hospital ill for the month he was in the hospital. I was not prepared for his death, his liver, kidneys, and finally his heart just stopped beating. Please help me find the reason for this terrible worse time opf my life. I have all the hospital records and he got acute bacterial infection at the end and could not fight this. He was on Avndia for 1 1/2 years 4 miligrama. I pray I can avenge his death due to this terrible medicine. Thank you, Sherrill Lynn Francis.
